【摘要】:Bacteriathatexistinbiofilmarephenotypicallydifferentfromsuspendedcellsofthesamegenotype.Researchhasshownthatbiofilmbacteriaaremoredifficulttokillthansuspendedbacteria(5).Laboratorybiofilmsareengineeredingrowthreactorsdesignedtoproduceaspecificbiofilmtype.Alteringsystemparameterswillcorrespondinglyresultinachangeinthebiofilm.Forexample,researchhasshownthatbiofilmgrownunderhighshearismoredifficulttokillthanbiofilmgrownunderlowshear(6).ThepurposeofthistestmethodistodirectauserinthelaboratorystudyofaPseudomonasaeruginosabiofilmbyclearlydefiningeachsystemparameter.Thistestmethodwillenableaninvestigatortogrow,sample,andanalyzeaPseudomonasaeruginosabiofilmgrownunderhighshear.ThebiofilmgeneratedintheCDCbiofilmreactorisalsosuitableforefficacytesting.Afterthe48hgrowthphaseiscomplete,theusermayaddthetreatmentinsituorharvestthecouponsandtreatthemindividually.1.1ThistestmethodspecifiestheoperationalparametersrequiredtogrowarepeatablePseudomonasaeruginosabiofilmunderhighshear(1).Theresultingbiofilmisrepresentativeofgeneralizedsituationswherebiofilmexistsunderhighshearratherthanrepresentativeofoneparticularenvironment.1.2ThistestmethodusestheCentersforDiseaseControlandPrevention(CDC)biofilmreactor.TheCDCbiofilmreactorisacontinuouslystirredflowreactorwithhighwallshear.AlthoughitwasoriginallydesignedtomodelapotablewatersystemfortheevaluationofLegionellapneumophila(2),thereactorisversatileandmayalsobeusedforgrowingand/orcharacterizingbiofilmofvaryingspecies(3and4).1.3Thistestmethoddescribeshowtosampleandanalyzebiofilmforviablecells.Biofilmpopulationdensityisrecordedaslogcolonyformingunitspersurfacearea.1.4Basicmicrobiologytrainingisrequiredtoperformthistestmethod.1.5ThevaluesstatedinSIunitsaretoberegardedasthestandard.Thevaluesgiveninparenthesesareforinformationonly.1.6Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
